COURSE SYLLABUS FOR BT115-SPRING 2013 INSTRUMENTATION FOR THE BIOTECHNOLOGY LABORATORY SPRING 2013 LECTURE HT405, FRIDAY 9 :30-11 :10AM LAB HT405, FRIDAY 11 :30AM-2 :10PM COURSE DESCRIPTION This course is a survey of the theory and practice of laboratory instrumentation with emphasis on biotechnology applications. Principles of measurement and calibration will be presented in the context of a GLP/cGMP environment. Quantitative analysis using UV-Vis spectrophotometry and fluorometric measurements will be described. Separation of biomolecules by chromatographic and electrophoretic methods will be presented. Laboratory robotics, Protein Sequencing, DNA Sequencing, and imaging techniques will be briefly introduced. PREREQUSITE: CH100A OR CONSENT OF DEPARTMENT OBJECTIVES: This course will introduce you to three of the fundamental instrumental methodologies used in the biotechnology, i.e. spectroscopy, chromatography, and mass spectrometry. Upon successful completion of the course you will understand the basic theory and operation of these instruments. You will have the ability to operate modern spectrophotometers and fluorometers for qualitative and quantitative measurements that are typically made in the biotechnology laboratory. You will be introduced to modern techniques and instrumentation for liquid and gas chromatography and mass spectrometry, will analyze chromatographic and mass spectrometric data, and will be introduced to the principles involved in sequencing proteins and DNA. STUDENTS WITH DISABILITIES: Any student who may need an accommodation due to a disability, please see me in private during office hours or by appointment as early as possible in the semester. A letter from Disability Support Services (SA175) will be needed. Any student who may need assistance in the event of an emergency evacuation must identify themselves to the Disability Support Services Office; guidelines for emergency evacuations for individuals with disabilities are found at: www.montgomerycollege.edu/dss/evacprocedures.htm. VETERANS AND ACTIVE RESERVISTS If you are a veteran or on active or reserve status and you are interested in information regarding opportunities, programs and/or services, please visit the Combat2College website at www.montgomerycollege.edu/combat2college and/or contact Joanna Starling at 240-567-7103 or [email protected]. OFFICE HOURS: After lab or by appointment GENERAL COURSE POLICIES 1. THE REQUIREMENTS OF THE DEPARTMENT AND PROGRAM AS SET FORTH IN THE “COMMON SYLLABUS” ALWAYS TAKE PRECEDENCE. ALTHOUGH THERE IS NO FURTHER SPECIFIC "CUT POLICY" FOR THIS COURSE, ATTENDANCE IN LECTURE AND LAB SESSIONS IS EXPECTED. 2. EYE PROTECTION MUST BE WORN WHEN WORKING IN THE LABORATORY. PRESCRIPTION GLASSES ARE ACCEPTABLE EYE PROTECTION. IF YOU DO NOT WEAR GLASSES, YOU MUST WEAR SAFETY GLASSES. DO NOT WEAR CONTACT LENSES WHILE WORKING IN THE LABORATORY! 3. THE GRADE YOU EARN WILL BE BASED ON TWO PROBLEM SETS, AND YOUR LABORATORY GRADE WHICH WILL CONSIST OF YOUR LABORATORY EXERCISES GRADE WHICH IS BASED ON YOUR LABORATORY NOTEBOOK AND YOUR LABORATORY PARTICIPATION. LABORATORY PARTICIPATION IS MY ASSESSMENT OF YOUR TECHNIQUE, PREPARATION, TEAMWORK, AND EFFORT. EACH ELEMENT WILL BE COMPRISED OF 100 POINTS, GIVING A TOTAL POSSIBLE OF 400 POINTS AND YOUR GRADE WILL BE BASED ON THE AVERAGE SCORE FOR ALL OF THE ELEMENTS. ALL WORK IS TAKE-HOME,OPEN-BOOK, OPEN-NOTES. USE WHATEVER RESOURCES YOU WANTLIBRARY, WEB SITES, WHATEVER. WORK TOGETHER IF YOU WANT, IT'S OK.

YOUR AVERAGE SCORE: (PROBLEM SET1 + PROBLEM SET2 + LABORATORY EXERCISES + LABORATORY PARTICIPATION) / 400) X 100

DEADLINES TO TURN IN WORK: YOUR MID-TERM GRADE WILL BE BASED ON YOUR SCORE ON PROBLEM SET I. PROBLEM SET I MUST BE TURNED IN ON OR BEFORE 2:10PM, MARCH 15, 2013. YOUR FINAL GRADE WILL BE BASED ON THE AVERAGE SCORE OF PROBLEM SET I (MID-TERM), PROBLEM SET II, YOUR LAB NOTEBOOK, AND YOUR LAB PARTICIPATION. PROBLEM SET II AND YOUR LAB NOTEBOOK MUST BE TURNED IN ON OR BEFORE 2:10PM, MAY 17, 2013. GRADE ASSIGNMENTS WILL BE MADE ACCORDING TO THE FOLLOWING SCALE: 90-100 = A 80-89 = B 70-79 = C LESS THAN 70 = YOU DON'T WANT TO BE HERE 4. THE RECOMMENDED TEXT FOR THIS COURSE IS: CHEMICAL ANALYSIS, Modern Instrumentation Methods and Techniques by Francis and Annick Rouessac, John Wiley & Sons, 2004ISBN 0-471-97261-4. HOWEVER, YOU MAY ELECT NOT TO PURCHASE THIS TEXT. YOU WILL RECEIVE ALL NECESSARY INSTRUCTIONAL MATERIALS IN CLASS OR VIA “MY COURSES”, THE COLLEGE WEB SITE FOR COURSE INFORMATION. THE TEXT ABOVE IS A FULL-SIZED TEXT THAT COVERS GENERAL ANALYTICAL CHEMISTRY TOPICS. THERE MAY BE MATERIAL DISCUSSED IN CLASS FOR WHICH THERE IS NO SPECIFIC HANDOUT OR CHAPTER, AND YES, YOU ARE RESPONSIBLE FOR THAT MATERIAL AS WELL. EVEN THOUGH THERE IS NO SPECIFIC TEXTBOOK, THERE ARE SEVERAL EXCELLENT RESOURCES IN ADDITION TO THE ONE ABOVE THAT YOU MAY FIND HELPFUL: “BASIC BIOSCIENCE LABORATORY TECHNIQUES” BY PHILIP BONNER AND ALAN HARGREAVES, ISBN#978-0-470-74309-6 THIS IS A POCKET-GUIDE THAT COVERS THE BASICS OF SEVERAL ESSENTIAL TECHNIQUES FOR BIOTECH LABS INCLUDING SPECTROPHOTOMETRY AND CHROMATOGRAPHY, ALTHOUGH IT DOES NOT COVER MASS SPECTROMETRY. ONE OF THE MOST ATTRACTIVE FEATURES OF THIS TEXT IS ITS AFFORDABILITY. "HANDBOOK OF INSTRUMENTAL TECHNIQUES FOR ANALYTICAL CHEMISTRY" BY FRANK SETTLE, ISBN#0-13-177338-0 "THE ESSENTIAL GUIDE TO ANALYTICAL CHEMISTRY" BY GEORG SCHWEDT, ISBN#0-471-97412-9 "BIOANALYTICAL CHEMISTRY" BY MIKKELSEN AND CORTON, ISBN#0-471-54447-7 "BASIC CALCULATIONS FOR CHEMICAL AND BIOLOGICAL ANALYSES" BY EFIOK ISBN#0-935584-51-X "ELECTRONICS AND INSTRUMENTATION FOR THE CLINICAL LABORATORY" BY EGGERT, ISBN#00471-86275-4 "INTRODUCTION TO FLUORESCENCE SPECTROSCOPY" BY SHARMA AND SCHULMAN ISBN#0-471-11098-1 "BASIC GAS CHROMATOGRAPHY" BY McNAIR AND MILLER, ISBN#0-0471-17260-X "MICROARRAY BIOCHIP TECHNOLOGY" EDITED BY MARK SCHENA, ISBN#1-881299-37-6 5. LAB NOTES, DATA, AND CALCULATIONS MUST BE RECORDED IN INK. THE KEY ELEMENT THAT DECIDES WHETER LABORATORY NOTES ARE GOOD OR WORTHLESS IS WHETHER ANYONE CAN PICK UP THE NOTES, UNDERSTAND WHAT YOU HAVE DONE AND RETRIEVE THE DATA THAT YOU HAVE OBTAINED.
