Thin Layer Chromatography (TLC) Analysis of Analgesic Drugs Reading

Experiment 12 in Pavia (5th edition pp 91-95). Read all of the required reading at the beginning of the experiment, especially Analgesics (p.75) and Identification of Drugs (p. 89).

Prelab Include the usual Name, Date, Title, Purpose, and Outline, plus find and draw in your notebook the structure of all of the analgesics that you will examine during this experiment (see below). Prelab Exercise Answer questions 1, 3, 5 and 6 on p. 822-823 (at the end of Technique 20) in Pavia. Turn these in on a separate sheet of paper! Procedure Follow the procedure in the experiment except for the following items: Although the instructor will demonstrate how to make a micro-pipette, one will be provided that you can use multiple times, you can clean the pipette after each use with acetone. The standard analgesic solutions will already be prepared. For the two reference plates (Ref. 1, and Ref. 2), use the following suggested scheme: Ref. 1: acetaminophen, caffeine, salicylamide, and the reference mixture Ref. 2: aspirin, ibuprofen, salicylamide and the reference mixture Perform analysis on two (2) known over-the-counter analgesic tablets plus at least one unknown tablet (see your instructor). Be sure to include the reference mixture on this plate, as well. Warning: You will be using a low power UV lamp that can cause damage to your eyes if you stare directly into the lamp. So don't stare into the lamp! To complete the experiment – Partial Report Put all measured Rf values in a Table. Sketch all TLC plates directly in your notebook, as accurately as possible. (you should be able to measure Rf values from your pictures in the notebook, if needed). Be sure to report the Rf of all observed components in the unknowns that you examine. Identify any components of the known and unknown analgesic tablets by comparing Rf values to known analgesics (from the reference plate). Answer Questions 1-5 on p. 95 (Optional exercise: perform an Internet search on any of the analgesics listed above and include in your conclusion a very brief history of it.) Version 20130925
